Mechano-Biology Based Optimal Design and Fabrication of Multi-Functional Bone Scaffolds

Healing large bone defects remains a challenge for clinicians and patients, primarily due to the limited availability of autologous bone grafts. Therefore, the use of scaffolds as carrier structures is a common practice. Among other functionalities, the scaffold should provide sufficient mechanical properties to endure physiological loading and offer enough porosity and surface area for nutrient and oxygen delivery, waste disposal, cell attachment, and bone growth. A proper design can be achieved by optimizing the bone scaffold’s geometry for sufficient strength, porosity, and material choice, while ensuring manufacturability. Therefore, the scope of my thesis proposes the design of bone scaffolds with optimal pore sizes, considering a mechanobiological model to maximize bone formation. Once the optimized bone scaffold geometry is obtained, the 3D printing process will be further optimized for the scaffolds. These scaffolds will then be used for cell culture to assess cell attachment and undergo mechanical characterization.
